In general, can you test an upgrade by testing it on your Delphix Replication engine? Can you smoothly run the replication from the lower version to the higher, and then cutover?

Unfortunately not, because Delphix Replication requires that both engines are running the same version. This is due to the fact that not just data, but also metadata (internal state) information must be replicated, and this can/will change between versions.

If you have more questions about replication please check out http://docs.delphix.com/display/DOCS40/Replication.

Also, we validate all upgrades in house to make sure that there is no issues for you and down time for upgrades is usually no more than 1-2 hours depending on the number of VDBs and dSources you have.
